Only 3 cylinders seem to run ( 3 of 4 exhaust outlet get hot) until the bike is warmed and you give it some decent throttle. Checked spark plug, its fine and gives good spark. Carburettor is clean and functional.

Is this a Compression issue? any ideas?

    Cheers

pilot jet circuit or enrichment circuit.

if the enrichment well in the bowls is clogged you will get no gas added when using choke lever.

pilot jet runs bike at low rpm maybe open mixture screw up a little more or the circuit is still clogged for that carb/cylinder.

compression: have you checked your valve clearances? do a compression test could be compression but would have to be quite off.

check the position of air jets on that carb and its fuel jets.
